There is an LED on the back of the changer labeled “Blocker Line 6”.  When I was testing the unit it and adjusting the switches, that light was always on?  Anyone know what it indicates? What it is blocking?

I have used them many times and I always thought it was a light saying it was getting power.

Basically the led is telling you that there is power to the vend circuit and that the coin mech should accept coins. If the led is off it will not accept coins. Line 6 on a coin mech is called the "crem" line the term blocker comes from the old style coin mechs that had the mechanical coils with metal fingers on the ends that would actuall protrude into the coins path and block it from being accepted.
